How anastrozole works
Many breast cancers are stimulated to grow by the female sex hormones oestrogen and progesterone. These breast cancers can be treated with drugs that block the effects of these hormones.
In women who have had their menopause, oestrogen is mainly produced by changing androgens (sex hormones produced by the adrenal glands) into oestrogens. This process is called aromatisation and happens mainly in the fatty tissues, muscle and the skin. It needs a particular enzyme called aromatase.
Arimidex blocks the process of aromatisation. So it lowers the amount of oestrogen in the body. In advanced breast cancer the cancer cells may grow more slowly or stop growing completely.




Usage in men
Anastrozole has been tested for reducing estrogens, including estradiol, in men. Excess estradiol in men can cause benign prostatic hyperplasia, gynecomastia, and symptoms of hypogonadism.
Some athletes and body builders use anastrozole as part of their steroid cycle to reduce and prevent symptoms of excess estrogen--gynecomastia, emotional lability and water retention.

ARIMIDEX (anastrozole) is a non-steroidal aromatase inhibitor. It is chemically described as 1,3-Benzenediacetonitrile, a, a, a', a'-tetramethyl-5-(1H-1,2,4-triazol-1-ylmethyl). Its molecular formula is C17H19N5.


Anastrozole is an off-white powder with a molecular weight of 293.4. Anastrozole has moderate aqueous solubility (0.5 mg/mL at 25°C); solubility is independent of pH in the physiological range. Anastrozole is freely soluble in methanol, acetone, ethanol, and tetrahydrofuran, and very soluble in acetonitrile.

DRUG CLASS AND MECHANISM


Anastrozole is an aromatase inhibitor, an anti-cancer medication use for the adjuvant or initial treatment of breast cancer in post-menopausal women. Estrogen causes or increases growth of certain breast cancers. Anastrozole works by blocking aromatase enzyme, which is involved in estrogen production in the body. This leads to decreased tumor size or delayed progression of tumor growth in some women. The FDA approved brand name anastrozole (Arimidex) in 1995.


PRESCRIBED FOR
Anastrozole is prescribed for hormone receptor-positive, early breast cancer in postmenopausal women as adjuvant treatment ( treatment following surgery with or without radiation ). Anastrozole is prescribed as a first-line treatment for hormone receptor-positive or hormone receptor unknown, locally advanced or metastatic breast cancer in postmenopausal women. Anastrozole is also prescribed for the treatment of advanced breast cancer in postmenopausal women with disease progression following treatment with tamoxifen (Nolvadex, Soltamox).

Other aromatase inhibitors include letrozole(Femara) and exemestane (Aromasin).


Many breast cancer tumors grow in response to estrogen. This medicine interferes with the production of estrogen in the body. As a result, the amount of estrogen that the tumor is exposed to is reduced, limiting the growth of the tumor.
