What is rofecoxib?

Rofecoxib (Vioxx) was withdrawn from the U.S. market in 2004.

Rofecoxib is in a class of dr-ugs called nonsteroidal anti-inflammatory d-rugs (NSAIDs). Rofecoxib works by reducing substances that cause inflammation, pain, and fever in the body.

Rofecoxib is used to reduce pain, inflammation, and stiffness caused by osteoarthritis, rheumatoid arthritis and certain forms of juvenile rheumatoid arthritis; to manage acute pain in adults; to treat migraines; and to treat menstrual pain.

Rofecoxib may also be used for purposes other than those listed in this medication guide.

Rofecoxib is a synthetic, nonsteroidal derivative of phenyl-furanone with antiinflammatory, antipyretic and analgesic properties and potential antineoplastic properties. Rofecoxib binds to and inhibits the enzyme cyclooxygenase-2 (COX-2), resulting in an inhibition of the conversion of arachidonic acid to prostaglandins. COX-related metabolic pathways may represent key regulators of cell proliferation and neo-angiogenesis. Some epithelial tumor cell types overexpress pro-angiogenic COX-2. (NCI04)

Rofecoxib is a nonsteroidal antiinflammatory drug (NSAID) that selectively inhibits cyclooxgenase-2 (Cox-2), which was used in the therapy of chronic arthritis and mild-to-moderate musculoskeletal pain. Rofecoxib was withdrawn in 2004 because of an association with an increase in cardiovascular events with its long term use. Rofecoxib had also been linked transient serum aminotransferase elevations during therapy and to rare instances of idiosyncratic drug induced liver disease.

How should I take rofecoxib?

Take rofecoxib exactly as directed by your doctor. If you do not understand these instructions, ask your pharmacist, nurse, or doctor to explain them to you.

Take each dose with a full glass of water.

Shake the rofecoxib suspension well before measuring a dose. Use a dose-measuring cup or spoon, not a regular table spoon, to measure the liquid form of rofecoxib to ensure that you measure the correct amount of medicine.
